Sofra Platform’s Breakthrough in Immune Activation

Noxopharm Limited (ASX:NOX) has revealed compelling preclinical data demonstrating that its Sofra™ platform can supercharge the immune system’s cancer-fighting arsenal. Central to this advance is Toll-like receptor 8 (TLR8), an immune sensor whose activation has been a hot target in oncology drug development. Noxopharm’s proprietary oligonucleotides have been shown to amplify TLR8 activity by over 200-fold in human skin biopsies ex vivo, a leap well beyond existing clinical-stage TLR8 agonists.

This enhanced activation was also observed in vivo, with a nearly three-fold boost in immune response in transgenic mice engineered to express human TLR8. These findings suggest a novel mechanism to intensify the immune system’s ability to attack cancer cells, potentially improving the efficacy of standard treatments like chemotherapy and radiotherapy.

The Sofra platform’s unique approach builds on recent breakthroughs in understanding innate immune regulation, as detailed in a publication in Nature Immunology and a related preprint. Noxopharm’s technology leverages short nucleic acid sequences, oligonucleotides, that modulate immune sensors to finely tune inflammation and immune activation.

Importantly, the company holds a granted US patent covering this technology, positioning it strategically in the burgeoning immuno-oncology landscape. The global market for cancer immunotherapies is forecast to surge from US$35 billion in 2025 to US$185 billion by 2035, underscoring the commercial potential of such innovations.

Advancing Toward Clinical Validation

With these promising preclinical results, Noxopharm is scaling up studies to assess the Sofra oligonucleotides in humanized TLR8 mouse models and various cancer models over the coming months. This progression aims to bridge the gap between laboratory findings and potential clinical application.

Professor Seija Lehnardt of Charité - Universitätsmedizin Berlin, an expert in TLR7/8 biology, remarked that the data indicate significant potential to enhance TLR8-targeted therapies. Meanwhile, CEO Dr Olivier Laczka highlighted that the Sofra platform’s deep mechanistic insights offer a distinct advantage in developing new immuno-oncology treatments.

This announcement follows Noxopharm’s recent efforts to refine its regulatory strategies for Sofra-based therapies, including engagements with global CRO Novotech to prepare for US FDA discussions on related indications regulatory strategy for SOF-SKN.
