Independent Validation Confirms AlgoraeOS v2 Advances

Algorae Pharmaceuticals (ASX:1AI) has taken a significant step forward in its AI-driven drug discovery efforts by completing an independent preclinical validation of its AlgoraeOS v2 (AOS2) platform. Conducted by the Victorian Centre for Functional Genomics at the Peter MacCallum Cancer Centre, the study tested 24 drug combinations with cannabidiol (CBD) across four cancer cell lines, generating an extensive dataset of around 10,000 data points.

The validation demonstrated that AOS2 substantially outperforms its predecessor, AOS1, by identifying roughly 2.5 times more confirmed synergistic drug combinations relative to predictions. This improvement underscores the enhanced predictive power of the updated platform, which integrates uncertainty metrics to better gauge the confidence of each prediction.

Robust Methodology Supports Predictive Accuracy

Employing high-throughput microscopy and chemiluminescent viability assays, the study assessed drug synergy using four established models: Highest Single Agent, Bliss independence, Zero Interaction Potency, and Loewe additivity. The close alignment of three of these models reinforced the consistency and robustness of the synergy predictions, while the uncertainty metrics added a valuable layer of precision in triaging candidate combinations.

These findings validate the iterative improvements made to AlgoraeOS, confirming its ability to prioritise drug combinations with a higher likelihood of true biological synergy. The company now plans to explore the most promising combinations further, including potential intellectual property protection.

Expanding the Pipeline with Multi-Anchor Combinations

Building on this momentum, Algorae is launching a multi-anchor combination program that expands its AI predictions from a single anchor drug to 18, evaluating over 9 million potential drug–drug–cell line combinations. This ambitious initiative has already identified more than 6,000 combinations with potential synergy, setting the stage for further in vitro validation of selected candidates.

The next phase is supported by a $49,109 voucher from Phenomics Australia’s Pipeline Accelerator, which will subsidise experimental work at the Peter MacCallum Cancer Centre. This funding follows a competitive selection process and reduces the cost burden on Algorae as it advances its drug combination discovery efforts.
