Immutep’s cash runway faces pressure after pivotal trial failure
Immutep’s pivotal TACTI-004 lung cancer trial dominates a difficult FY26, with the failure widening the loss to A$79.3 million and triggering an impairment, cost cuts and a root cause review. The biotech retains A$68.9 million in cash, cash equivalents and term deposits, while its remaining investment case rests on IMP761 and selected efti studies.

TACTI-004 failure dominates Immutep’s FY26
Immutep Limited (ASX:IMM; NASDAQ:IMMP) spent FY26 advancing a broad immunotherapy pipeline, but the year was ultimately defined by the collapse of its most important trial. The company discontinued TACTI-004, a Phase III study testing eftilagimod alfa, or efti, with pembrolizumab and chemotherapy in first-line non-small cell lung cancer, after an interim futility analysis conducted on approximately 170 patients.
The Independent Data Monitoring Committee recommended stopping the study in March 2026, and Immutep accepted the recommendation after reviewing early progression-free survival data. The company said patients receiving the efti combination were overall underperforming the control arm, an outcome it described as unexpected given earlier results from similar treatment combinations. Enrolment was halted and the study entered an orderly wind-down, with follow-up and site close-out activities continuing.
The company’s root cause analysis remains in progress across clinical, operational, analytical and manufacturing factors. That review now sits at the centre of Immutep’s next decision on efti, following the manufacturing differences review, which earlier coverage said had identified differences between batches made at different scales.
Loss and impairment expose the financial cost
Immutep’s net loss widened to A$79.3 million from A$61.4 million, while research and development and intellectual property expenses rose to A$91.9 million from A$61.4 million. The increase reflected heavier clinical activity, TACTI-004 close-out costs and the root cause investigation. A further A$3.1 million non-cash impairment was recognised against efti-related intellectual property.
Revenue and other income nevertheless more than doubled to A$24.2 million, from A$10.3 million, helped by A$14.6 million of licensing revenue recognised from the Dr Reddy’s Laboratories agreement. Immutep received a US$20 million upfront payment in January, but repaid US$10 million in June after TACTI-004 was discontinued. The remaining commercial terms, including potential milestone payments of up to US$349.5 million and royalties in licensed territories, were unchanged according to the report.
Cash preservation becomes the immediate strategy
At 30 June, Immutep held A$63.7 million in cash and cash equivalents plus A$5.2 million in term deposits. Operating cash outflow was A$55.5 million, an improvement on the A$62.0 million outflow recorded in FY25, although the comparison is complicated by licensing receipts and the timing of payments. Management said headcount reductions, lower operating expenditure and the TACTI-004 wind-down should materially reduce future cash outflows, forecasting a cash runway into the first half of CY2028 under current forecasts.
That runway is not the same as commercial self-sufficiency. The accounts note recurring losses, negative operating cash flows and the possibility that Immutep may need future licensing transactions, capital raisings or tighter control of research spending. Nasdaq also notified the company in April that its American Depositary Shares had fallen below the US$1 minimum bid-price requirement, with an initial compliance period expiring on 26 October 2026; the annual report does not state the outcome of that process.
Efti retains pockets of clinical support
Other efti programs produced more encouraging signals, though none carries the evidentiary weight of a successful registrational trial. Investigator-led INSIGHT-003 reported median overall survival of 30.9 months in first-line non-squamous NSCLC, compared with a 22.0-month historical benchmark. The comparison was against a previous third-party trial rather than a randomised control arm, and the company cautioned that the study population had a higher proportion of patients with low PD-L1 expression. Earlier coverage also highlighted the divergence between INSIGHT-003 and TACTI-004 in lung cancer outcomes.
EFTISARC-NEO, a Phase II investigator-initiated study in soft tissue sarcoma, met its primary endpoint and reported a median tumour hyalinisation or fibrosis rate of 51.5% among 38 evaluable patients, above the prespecified 35% target and historical results of about 15% for radiotherapy alone. The FDA granted efti Orphan Drug Designation in April. Immutep has also received FDA feedback supporting further development in PD-L1-negative head and neck cancer, while the AIPAC-003 metastatic breast cancer study closed at the end of June after the FDA agreed that 30 mg was the optimal biological dose under Project Optimus.
IMP761 offers the clearest separate development path
Immutep’s autoimmune program may now carry greater strategic importance. IMP761, described as a first-in-class LAG-3 agonist antibody, completed the single ascending dose phase of its Phase I trial with favourable safety and pharmacodynamic findings. Through doses of up to 14 mg/kg, the company reported no serious treatment-related adverse events and said the drug substantially inhibited T-cell infiltration after an antigen challenge in vaccinated participants. The multiple ascending dose phase was under way at year end, extending the development path previously outlined in coverage of the program.
The board also reported 19 new patents across efti, IMP761 and LAG525 during the year. Novartis’ licence and collaboration agreement covering LAG525 was terminated in August, with the associated intellectual property to be assigned to Immutep; the company said the termination had no material financial impact. Pete Meyers, a non-executive director and deputy chairman, is listed as having resigned on 2 October 2026, after the reporting period.
Immutep enters FY27 with enough stated liquidity to buy time, but not enough certainty to remove the central question: whether the root cause analysis can distinguish a trial-specific failure from a problem that limits efti’s wider development case. Until that answer arrives, IMP761 and the smaller efti studies must carry a larger share of the company’s future narrative.
Bottom Line?
Immutep has bought development time through cost reductions and licensing income, but the value of that runway depends on what the TACTI-004 review says about efti’s future.
Questions in the middle?
- Can the root cause analysis explain why TACTI-004 diverged from earlier efti studies, including INSIGHT-003?
- Will management commit capital to new efti trials, or concentrate resources on IMP761 and narrower indications?
- How quickly will the lower-cost operating model translate into reduced cash burn and extend the stated runway?
