PYC Therapeutics has moved all four RNA drug programs towards their next development milestones, while reporting $655 million in cash and treasury investments at 30 September. The company’s standalone cash balance covers just 1.13 quarters, making its liquid investment portfolio central to the stated funding runway through 2030.
- PYC-003 advanced to 2.4 mg/kg dosing and a 24-month open-label extension in PKD
- PYC-002 entered GLP toxicology studies ahead of expected human trials in 2027
- PYC reported clinical or pre-clinical data updates for ADOA and RP11 programs
- $655 million held across cash, term deposits and liquid financial assets
- Cash and cash equivalents alone covered 1.13 quarters of operating outflows
Four Programs Move Towards Clinical Milestones
PYC Therapeutics Limited (ASX:PYC) has advanced all four of its genetic disease programs, with the most consequential near-term work now concentrated in repeat-dose clinical studies, toxicology and proof-of-concept readouts. The update is encouraging in breadth, but much of the pipeline remains early-stage and subject to trial outcomes, regulatory alignment and other disclosed risks.
In polycystic kidney disease, PYC-003 progressed to dosing at 2.4 mg/kg in the Phase 1b Part C multiple ascending dose study after safety review of earlier data. The company also began an open-label extension allowing eligible participants to receive treatment for up to 24 months. That builds on the earlier PKD dose escalation, which allowed dosing to increase and introduced the extension study.
PKD Data Remains the Near-Term Clinical Test
PYC said completed single-dose cohorts showed no treatment-related serious adverse events and no safety-related changes in liver or kidney function. It has also presented early exploratory efficacy data from PKD patients, although those findings do not yet establish clinical proof of concept. Enrolment is being completed in the repeat-dose and open-label studies, with data expected in calendar 2027.
The other programs are at different points on the development curve. PYC-002 for Phelan-McDermid Syndrome has entered the GLP toxicology work required before clinical trials, while patient-derived models showed maintained SHANK3 isoform balance and restored neuronal communication after treatment. PYC expects to progress the candidate into human studies in 2027, subject to regulatory alignment.
Eye Disease Candidates Produce Further Signals
For autosomal dominant optic atrophy, PYC presented updated Phase 1/2 data showing improvements in low-contrast visual acuity and reduced macular stress after repeat dosing. The company also reported non-human primate data showing increased target gene expression in the retina four months after a single dose, supporting its evaluation of dosing intervals longer than four months. The update extends the earlier ADOA vision data, which reported sustained visual and retinal improvements.
In retinitis pigmentosa type 11, PYC presented updated long-term safety and efficacy data from the ongoing Phase 1/2 program for VP-001 and is using those data to inform a potential registrational study. Across both eye-disease programs, the company’s next meaningful readouts are expected in 2027, but the filing does not provide the participant numbers or effect sizes needed to assess the strength of the reported signals independently.
Liquidity Is Stronger Than the Cash-Only Runway Suggests
PYC reported $655 million in cash and treasury investments at 30 September, comprising $17 million in cash and cash equivalents, $200 million in term deposits and $438 million in floating-rate notes and US Treasury bills. Quarterly operating cash outflow was $14.9 million, after $21.4 million of research and development payments and $8.1 million of interest received.
On the narrow Appendix 4C measure, cash and cash equivalents alone represented 1.13 quarters of funding. PYC said that measure excludes investments held to fund operations and stated that its broader funding position supports development of all four programs through calendar 2030. It also said it does not currently propose a specific capital raise, while retaining the option to consider funding alternatives over time.
Bottom Line?
The pipeline now has several 2027 decision points, but the investment case turns on whether early clinical signals become reproducible proof of concept before the treasury portfolio begins carrying a heavier funding burden.
Questions in the middle?
- Will repeat-dose PKD data demonstrate proof of mechanism and proof of concept in 2027?
- Can the reported ADOA and RP11 signals translate into registrational-quality evidence?
- How quickly will PYC need to convert term deposits and liquid investments into operating cash as development spending rises?
